HBV Extraction Kit “HBV Extraction Kit is IVD”
Overview The HBV Extraction Kit is an automatic extraction system for the isolation of high -quality DNA from serum/plasma. This Extraction Kit is used with the Nextractor® to provide high-yield and quality DNA from your sample. Please refer to the manual for details about the Nextractor®.
Operation Principle Transfer the specimen into the sample well, insert the plate into the Nextractor® and start the program. The first step of the extraction is 『lysis and binding DNA to magnetic beads』, in which the specimen is mixed with lysis solution and magnetic beads. In this step, DNA is extracted from the specimen and then bound to magnetic beads. The second step of the extraction is 『DNA Wash Ⅰ/ Wash Ⅱ』. In this step, inhibitors affecting the PCR reaction are removed by washing solution. The third step of the extraction is 『DNA Elution』. In this step, DNA attached to the magnetic beads is dissolved into the elution buffer.

Warning and Precautions
1. All samples must be treated as potential biohazards. 2. Wear appropriate protective eyewear, clothing, and gloves. 3. Avoid all skin contact with kit reagents. In case of contact, wash thoroughly with water. 4. Minimize the inhalation of chemicals. Do not leave chemical containers open. Use only with adequate ventilation (i.e. a fume hood). 5. All work should be conducted in properly equipped facilities using the appropriate safety equipment (i.e. physical containment devices). 6. Individuals should be trained according to applicable regulatory and company/institutional requirements before working with potentially infectious materials.

Experimental Methods The HBV Extraction Kit should be used with the Nextractor®(Gyeongin 14-66). ※ All extractions must be processed in 2~35℃. ※ Our extraction kit must be stored in 2~35℃. If the extraction kit has been stored above 35℃, the result would be affected. 1. Sample Collection and Storage 1.1 Sample collection Collect samples in a sterile container. 1.2 Sample storage 1) Store at 4°C for up to one week. 2) Store at –70°C for long-term storage. 2. Specimen Preparation 2.1 Serum/Plasma samples 1) Vortex Serum/Plasma samples for 10~30 sec. ※ If samples are stored frozen, submerge them in a water bath at 25–37°C until completely thawed, then place on ice until further use. 3. Experimental methods HBV DNA extraction from serum/plasma sample (Cat No. HB1102, HB1111) 1. Determine the number of samples to be tested and carefully remove the sealing tape from the cartridge of the HBV Extraction Kit. 2. Add 200μl of serum or plasma into the sample wells(96-well : 1st, 5th, 9th well, 32-well : 1st well) of HBV Extraction plate. 3. Insert the plate and strip to Nextractor®and start Universal for Extraction. 4. Once the operation is done, take out the plate and take 5 μl DNA for HBV detection. - If there is the magnetic bead in the Elution Buffer after extraction, take the
DNA/RNA after placing the ‘well plate’ on top of the provided ‘magnetic board’ so that the ‘magnetic beads’ are fixed to the magnetic board.
