Meaning of the Symbols Used on the Product LED safety The light in the near-infrared region (IR wavelength) emitted from the infrared LED is applied to the focusing control. This product conforms to the EU standard EN60825-1: 2001 and the international standard IEC60825-1: 2001. Additionally, the LED safety is classified to Class 1. • The safety label is attached on the upper surface of the LV-DAF intermediated tube and the safety standard/rating label is on the side of the LV-DAF controller. • For fear of exposure to radiation in the light from the LED, never attempt to operate or adjust the product according to the instructions that are not indicated in this manual. Also, do not remove the cover from the product. • Normally, IR light is emitted from the attachment location of the lower surface of the intermediate tube. Do not use the product as a single unit. Be sure to use it with assembled to the microscope system. Normally, IR light is not emitted from the eyepiece tube mounting location on the upper surface of the intermediate tube. Do not touch when carrying the product. The symbols on the both sides of the LV-FMA motorized focusing module elevating section call your attention on the following. For positions of this symbol, refer to the instruction manual of the LV-FMA. • Do not touch the elevating section and arm when carrying the product. 3 WARNING 1. Intended product use The product is an equipment to perform the auto focus function (automatic focusing) with assembled to the Nikon LV series microscope. Do not use the product for other purposes. 2. Do not disassemble. Disassemble of the product may result in electric shock or malfunctions. Never attempt to disassemble any part other than the parts described in this manual. If you notice abnormalities in the product, contact your nearest Nikon representative. 3. Read the instructions carefully. To ensure safety, carefully read this manual and the manuals for other equipment used with this microscope. In particular, observe all warnings and cautions given in the first of each manual. 4. Rating of the power supply The power supply circuit in the product is designed for AC power of 100 to 240 VAC and 50/60 Hz. Before connecting the power cord, check that the power supply to be used conforms to the voltage and frequency described above. Use of a non-conforming power line may result in equipment malfunction, failure, or fire. 5. Power cord Be sure to use the specified power cord. Use of a wrong power cord may result in malfunctions or fire. Additionally, be sure to connect the product to an appropriate ground terminal (protective earth terminal) because it is classified as subject to Class I protection against electric shock. To prevent electrical shock, always turn off the power switch (press the “ ” side) before attaching or detaching the power cord. Refer to “IX. Specifications” for the specified power cord. 6. Object devices of the product The product can perform the auto focus function if assembled with the following devices. The product cannot perform the auto focus function if assembled with the others. • Motorized focusing module: LV-FMA FM module A, LV-IMA IM module A • Motorized nosepiece: LV-NU5A U5A nosepiece, LV-NU5AC U5AC nosepiece • PC: Windows PC with a USB interface or Windows PC with an RS232C interface 7. Motorized devices operations The motorized devices (motorized focusing module and motorized nosepiece) can be operated by controlling the product. To prevent injury, do not touch the motorized devices during operation. Before operating the product, make sure that the motorized devices can be operated safety. Particularly, if you touch the elevating section of the motorized focusing module or the devices attached to the elevating section during performing the auto focus function, it may cause injury to hands or fingers. Do not touch the devices when you operate the LV-DAF. 8. Heat from the light source The lamp and the lamp house become extremely hot. To avoid burns, do not touch the lamp house while the lamp is lit or for thirty minutes after it is turned off. Additionally, to avoid the risk of fire, do not place fabric, paper, or highly flammable volatile materials (such as gasoline, petroleum benzine, paint thinner, or alcohol) near the lamp house while the lamp is lit or for about thirty minutes after it is turned off. 9. Cooling fan and air vents Do not block the opening for the cooling fan on the back and air vents on the side. If the cooling fan or air vents are blocked, the inside temperature of the product rises and it results in failure, malfunction, or fire. 4 CAUTION 1. Handle carefully. Handle the product carefully to avoid shock or impact. • Do not forcefully pull or twist the power cord and the cables. • Do not drop metal pieces such as clips and staples into any clearance or the product inside. It may result in failures or fire. 2. Do not wet the product. If the product gets wet, it may cause a short circuit and result in failure or abnormal heating. If the product accidentally gets wet, immediately turn off the power switch (press the “ ” side) and unplug the power cord from the wall outlet. Then, wipe off water with a piece of dry cloth. If water enters the product, immediately suspend use of the product and contact your nearest Nikon representative. 3. Weak electromagnetic wave The product emits weak electromagnetic waves. Do not place any precision electronic devices near the product to prevent its accuracy from being adversely affected. If a TV or radio reception is affected, move the TV or radio set farther from the product. 4. Installation location The product is a precision optical instrument. Therefore, use or storage of the product in an inappropriate environment may result in failures or poor performance. Consider the following requirements when arranging an installation location. • Avoid bright locations where the product is exposed to direct sunlight or immediately under room lights. If the ambient light is bright, the image will deteriorate. • Install the product with a surrounding clearance of 10 cm or more. • Install the product in a location that is free from considerable dust or dirt. • Install the product on a flat surface with little vibration. • Install the product on a sturdy desk or table to avoid breakage of the product. • Do not install the product in a hot and humid location. • Install the product in a location where the power cord can be easily unplugged from the AC inlet in the event of an emergency. • Refer to “IX. Specifications” for operation and storage conditions. 5. Cautions on assembling the product • Be sure to turn off the power switch before assembling the product or attaching/detaching the cable. • Be careful not to pinch your fingers or hands during assembly. • Scratches or fingerprints on the lens will adversely affect the image. Be careful not to scratch the lens during assembly. 6. Cable routing Route the cables away from the light source and lamp house. If a cable comes into contact with the lamp house, the cable sheath may melt and it results in an electrical shock or fire. 7. Setup works after assembly Perform setup works for the product when assembling the microscope system or changing the configuration. In the setup work, register information of the objective attached to the motorized nosepiece and specify the focus position of each objective. Be sure to perform the setup work before operation to perform the auto focus function correctly. The product is controlled to focus on specimens by projecting the slit image to be used for focusing on the specimen using IR light (near-infrared light) and detecting its reflection. The focus position is controlled by the hybrid method of the slit projection method and the slit image contrast detection method so that the focus range can be enlarged and the auto focus function can be performed for both diascopic illumination and episcopic illumination. To perform the auto focus function using this product, the following motorized devices must be assembled to the microscope system. • Motorized focusing module (LV-FMA or LV-IMA) • Motorized nosepiece (LV-NU5A or LV-NU5AC) This product can be assembled to the motorized microscope system accommodating the LV-ECON E controller. When using the LV-ECON, the motorized devices operations other than focus function is performed with the LV-ECON and the auto focus and manual focus functions are performed with the controller of this product. A fixing clamp screw is attached in the front of this device. 2 Mounting part (circular dovetail joint) This is the circular dovetail joint to attach this device onto another device. 3 Slot for analyzer slider An analyzer slider is inserted when performing the polarization microscopy or DIC microscopy. The indicator shows the focusing conditions. 8 OBJ. control panel It is used to rotate the motorized nosepiece to change objectives. 9 AF OFFSET control panel It is used to move the elevating section of the motorized focusing module and change the focus position under the auto focus condition. 10 FOCUS control panel It is used to move the elevating section of the motorized focusing module when the auto focus function is turned off or when the auto focus function is turned on but the DISABLE indicator is lit. 11 SEARCH control panel It is used to search focus positions. 12 MIC button It is used to set the microscopy method to be performed. 13 AUX. control panel Not used. 14 Tilt adjustment legs They are used to adjust the tilt angle of the product. 9 II. Normally, do not remove the cover. 16 USB connector It connects the devices such as the PC for setup work or the PC for external control. (This interface complies with USB 1.1.) 17 SENSOR connector It connects with the LV-DAF intermediate tube using the provided connecting cable. 18 SERIAL connector It connects the devices such as the PC for setup work or the PC for external control. When used with the motorized microscope system accommodating the LV-ECON, use this connector to connect this device and the LV-ECON. 19 I/O connector It is used for digital input and out control. 20 LV-FMA connector It is used to connect the motorized focusing module (Nikon LV-FMA or LV-IMA). 21 NOSEPIECE connector It is used to connect the motorized nosepiece (Nikon LV-NU5A or LV-NU5AC). When used with the motorized microscope system accommodating the LV-ECON, do not connect the motorized nosepiece to this connector. Connect the motorized nosepiece to the LV-ECON. 22 Power switch This is the power switch of this product. 23 AC inlet It is used to connect to the AC power supply. Be sure to use the specified power cord. 24 Cooling fan This is the cooling fan of this product. Assembly The procedures for assembling and connecting the product are described in this chapter. Refer to the instructions in this chapter for connection of the devices. Refer to the instruction manual of each device for installation and assembly. WARNING • Before connecting devices, be sure to read the WARNING and CAUTION in the first of this manual and follow the instructions written therein. • To avoid electrical shock or fire, be sure to turn off the power of the product (press the “ ” side of the power switch) before unplugging the power cord. CAUTION • Be careful not to pinch your fingers or hands during assembly. • Scratches or fingerprints on the lens will adversely affect the image. Be careful not to scratch the lens during assembly. If lenses are contaminated with fingerprint or such, clean them according to the procedure described in “VIII. Care and Maintenance”. • This product is a precision optical instrument. Handle the product carefully and do not subject it to a strong physical shock. Especially, the accuracy of the objective may be adversely affected by even a slight damage. • Route the cables away from the illuminator or lamp house. If a cable comes into contact with the lamp house, the cable sheath may melt and it results in an electrical shock or fire. CAUTION • Setup work for the “Auto Focus” Perform the setup work for the product after assembling the microscope or changing the configuration. In the setup work, register information of the objective attached to the motorized nosepiece and specify the focus position of each objective. Make sure to perform the setup work before operation to perform the auto focus function correctly. (This device needs a power supply for its own.) 13 2 Attaching the Intermediate Tube Attach the intermediate tube of the LV-DAF to the microscope system. The procedure is as follows: 1 Check that the motorized focusing module (LV-FMA or LV-IMA) is secured to the installation position. 2 Attach the motorized nosepiece (LV-NU5A or LV-NU5AC) to the motorized focusing module. Refer to the instruction manual of the LV-FMA or LV-IMA for details about the motorized nosepiece attachment method. 3 To use the episcopic illuminator, mount it on the arm of the motorized focusing module. 4 When using the episcopic illuminator, mount the intermediate tube of the LV-DAF on the episcopic illuminator and secure it using a clamp screw. When not using the episcopic illuminator, mount the intermediate tube of the LV-DAF on the arm of the motorized focusing module and secure it using a clamp screw. 5 Attach the eyepiece tube to the upper side of the intermediate tube of the LV-DAF and secure it using a clamp screw. 6 Attach objectives to the motorized nosepiece so that the magnification increases as the nosepiece is rotated clockwise as viewed from above. Assembly 3 Attaching the IR Cut Lens or IR Cut Filter The product controls the focusing conditions using the near-infrared (IR) light. Therefore, to operate the product correctly, IR light needs to be removed from the illumination. Attach the IR cut lens or IR cut filter as follows: For episcopic illumination To perform episcopic illumination with the episcopic illuminator, attach the provided IR cut lens to the lamp house connector part of the illuminator. WARNING In the following procedure, the lamp house is attached or detached. To prevent the risk of burns, make sure that the lamp is turned off and its surrounding area are cooled down sufficiently before operation. 1 When the lamp house is attached to the microscope system, make sure that the lamp is turned off and the lamp house is cooled down sufficiently. Then, remove the lamp cable from the lamp house and loosen the clamp screw to remove the lamp house. 2 Insert the IR cut lens into the lamp house connector part of the episcopic illuminator (LV-UEPI, LV-UEPI2, or LV-UEPI2A). Insert the IR cut lens as far as it goes with the lens facing inside of the microscope. 3 Attach the lamp house and secure it using IR cut lens a clamp screw. 4 Connect the lamp cable to the power supply. Do not touch the height adjustment par t when carrying this product. IR cut lens Lamp house Refer to the instruction manual of the episcopic illuminator (LV-UEPI, LV-UEPI2, or LVUEPI2A) for details about installation method of the lamp house and replacement method of lamps. For diascopic illumination IR cut filter To perform diascopic illumination with the LV-DIA DIA base, attach the optional IR cut filter on the field lens of the LV-DIA. F.S. 15 4 Connecting the Controller Connect the intermediate tube of the LV-DAF, the motorized focusing module, and the motorized nosepiece to the controller of the LV-DAF using the special connecting cables. 1 Make sure that the power switch on the back of the LV-DAF controller is turned off (the “ ” side is pressed down). 2 Connect the connector on the side of the intermediate tube and the SENSOR connector on the back of the controller using the special connecting cable. 3 Connect the connector on the side of the motorized focusing module and the LV-FMA connector on the back of the controller using the special connecting cable. 4 Connect the cable of the motorized nosepiece with the NOSEPIECE connector on the back of the controller using the special nosepiece connecting cable. An optional connection cable with a length of 10 m is also available. 16 III. Assembly 5 Connecting PCs Connecting a PC for the setup work To ensure correct operation, specify the information of objectives via a PC according to the special setup software “LV-DAF Setup.” Use the RS232C interface or USB interface to connect the product to the PC. After assembling and connecting the product, connect the PC with the controller using the provided RS232C cable (crosswired type cable) or USB cable. And then, activate the “LV-DAF Setup” to specify the information of objectives. Refer to “ECLIPSE LV Series Support Tools Software manual” in the provided CD-ROM for details about specifying the information of objectives. • For the RS232C interface application LV-DAF controller SENSOR SERIAL I/O LV-FMA USB 100-240V~ 1.2A 50/60Hz NOSEPIECE PC RS232C cable (cross-wired type cable) • For the USB interface application SENSOR SERIAL I/O LV-FMA USB 100-240V~ 1.2A 50/60Hz PC NOSEPIECE LV-DAF controller USB cable * The USB interface of the product is compatible with USB 1.1. 17 Connecting a PC or other device for external control The product can be controlled with a PC or other device using the USB interface or RS232C interface. Assembly 6 System Configuration with the LV-ECON The LV-ECON E controller is the controller for operating all motorized devices (LV series). When applying the product to the motorized microscope system accommodating the LV-ECON, change the connection as follows: • Connect the motorized nosepiece to the LV-ECON. • Connect the LV-ECON and the LV-DAF controller using the RS232C interface. • Connect the LV-ECON to the PC or DS-L2 using the USB interface. Refer to the following diagram for connection. Assembly 7 Connecting the Power Cord WARNING Be sure to use the specified power cord. Use of a wrong power cord may result in malfunction or fire. Additionally, be sure to connect the product to earth ground (protective earth terminal) because it is classified as Class I protection against electric shock. Refer to “IX. Specifications” for details of the power cord. Make sure that the power switch is turned off (the “ ” side is pressed down). Insert the socket into the AC inlet on the back of the device. Then, securely plug the power cord to a wall outlet. 8 Tilt Adjustment Legs The tilt adjustment legs for changing the tilt angle of the product are attached to the front legs part of the product. Check the installation condition of the product, and then use the tilt adjustment legs if you wish. 20 Setup Works The LV-DAF Dynamic auto focus system must be set up after the following cases: The system is used for the first time. The objective configuration is changed in the microscope. The specimen is changed. The controller is changed. Or, the intermediate tube is changed. To perform the setup works, use the provided software “LV-DAF Setup” on a PC. Refer to the “ECLIPSE LV Series Support Tools Software manual” in the provided CD-ROM for details about “LV-DAF Setup” operation and setup works. CAUTION If the setup work is not performed, this product does not work correctly. Be sure to set up the product. 21 Operation WARNING 1 Each motorized device will move when the product is operated. To prevent injury, do not touch the motorized devices during operation. Before operating the product, make sure that the motorized devices can be operated safety. Power On/Off Power On/Off The power switch is located beside the AC inlet on the rear. To turn on the product, press the “ I ” side of the power switch. To turn off the product, press the “ ” side of the power switch. When the product is turned on, the POWER indicator on the front of the product lights green. • Initialization of the motorized devices When the product is turned on, a beep sounds, all indicators on the operation panel turn off, and the product starts communications with each motorized device. When the communication is established, each motorized device is set to the predetermined conditions and the indicators on the operation panel show the current condition. This procedure is called the “initialization” of the microscope system. When the initialization ends, a beep sounds again to inform that the product is ready for use. Operation 2 Selecting the Microscopy Method To perform the auto focus function, the setting of the product must be changed suitable for the microscopy method. The DISABLE indicator is lit when this system cannot focus on the specimen using the auto focus function. To turn off the auto focus function, press the ON button and turn off the indicator. 23 4 Selecting an Objective To select an objective, press the + / – button of the OBJ. control panel to rotate the motorized nosepiece. The “1” to “5” indicators of the OBJ. control panel show the nosepiece address of the objective in the optical path. Operation 5 AF Offset Function After focusing on the specimen with the auto focus function, focal position can be shifted with remained in the auto focus function. This operation is called “AF offset”. The offset value of each magnification (each address of the motorized nosepiece) can be registered. To perform the “AF offset” function, press the ON button of the AF OFFSET control panel and light up the indicator after focusing on the specimen with the auto focus function. Operation of the “AF OFFSET” On/Off Then, operate + / – button of the AF OFFSET control panel to move the elevating section of the motorized focusing module so that the focal position can be moved. Operation Elevating Section Objective + button Up The objective moves farther from the specimen. – button Down The objective moves closer to the specimen. To turn off the auto focus function, press the ON button again and turn off the indicator. When the “AF offset” function is turned off, the offset values are reset. Note the followings: When the indicator is lit and the specimen is in focus, the AF offset function is running. When the AF offset function is running, the performance of auto focus will be less good than the normal condition. 25 6 Search Function If the specimen does not come into focus with the auto focus function after manual focusing or such operation, execute the search function to search a focusing position automatically. The search function has “Single mode” and “Continuous mode.” Execution of the search function Single mode On this mode, the search function ends when a focusing position is found. To invoke the single mode, press the ON button in the SEARCH control panel while the DISABLE indicator in the AUTO FOCUS control panel is lit. The indicator in the SEARCH control panel turns on. When the search function starts, the motorized focusing module moves upward and searches a focusing position. When a focusing position is found, the search function ends. If no focusing position is found, the DISABLE indicator in the AUTO FOCUS control panel turns on. Continuous mode On this mode, the auto focus function starts automatically when a focusing position is found. To invoke the continuous mode, press the ON button in the AUTO FOCUS control panel while the DISABLE indicator in the AUTO FOCUS control panel is lit. The AF indicator in the AUTO FOCUS control panel turns on. And then, press the ON button in the SEARCH control panel. The indicator in the SEARCH control panel turns on. When the search function starts, the motorized focusing module moves upward and searches a focusing position. When a focusing position is found, the auto focus function automatically starts. If no focusing position is found, the DISABLE indicator in the AUTO FOCUS control panel turns on. In the initial setting, the motorized focusing module moves only in the upward direction. But if the focusing position cannot be searched even when the module moves to the upper limit position, the module can be set to move in the downward direction. Additionally, when the module is set to move in the downward direction, be careful of a collision of the objective with the specimen. To prevent the collision, set up the software limit with “LV-DAF Setup” in the provided CD-ROM. 26 V. Operation Manual Focusing Operation of the manual focusing By operating the + / – button of the FOCUS control panel, the indicator is lit and then the elevating section of the motorized focusing module can be moved manually. To change the focusing mode between the fine focusing and the coarse focusing, press the FINE buttom. In the fine focusing mode, the FINE indicator turns on. If any of the troubles listed in the table below arise, take the countermeasures indicated. Trouble Cause Countermeasure Unable to turn on the product. The power cord is not connected. Connect it properly. (p.20) Unable to focus on the specimen by the auto focus function. Specimens other than the one to be detected are observed. Observe the transmission or nontransmission specimens with the boundary surface whose reflectance is 4% or more. Specimen is out of the focusing available range. (p.11 to 20) Care and Maintenance Nikon recommends daily care and maintenance for maintaining the performance as long as possible. Do not let dust, fingerprint, etc. get on the lenses. Soil on the lens or filter adversely affects the optical performance of the microscope. If lenses are contaminated, clean them according to the procedure described in “1. Cleaning Lenses and Filters.” When cleaning, be sure to turn off the power switch of the microscope (press the “ ” side) to avoid malfunction. Daily care and maintenance Clean the parts frequently manipulated by hands, such as eyepieces and glass plate, according to the procedure described in “1. Cleaning Lenses and Filters” without removing them from the microscope. Nikon recommends cleaning them frequently. Clean the tip of the objectives and filters to maintain the optical performance. When cleaning the objectives, remove them from the microscope. Clean them whenever they are contaminated. Microscopes and stages are contaminated with use. When you find the microscope is contaminated, clean them according to the description in “2. Care and Maintenance 1 Cleaning Lenses and Filters Do not let dust, fingerprint, and so on get on the lenses and filters. Soil on the lens or filter adversely affects the viewing of the image. If any of the lenses get dirty, clean them as described below. • Either brush away dust with a soft brush, or else gently wipe it off with a piece of gauze. • Only if there are fingerprints or grease, dampen a piece of soft, clean cotton cloth, lens tissue, or gauze lightly with absolute alcohol (ethyl or methyl) and gently wipe off the soil. • Absolute alcohol is highly flammable. Be careful when handling it, when around open flames, when turning the power switch on/off, and so on. • Follow the instructions provided by the manufacturer when using absolute alcohol. 2 Cleaning the Painted Parts, Plastic Parts, and Printed Parts Do not use organic solvents such as alcohol, ether, or paint thinner on painted components and plastic components. Doing so could result in discoloration or in peeling of the printed characters. For persistent soil, dampen a piece of gauze with neutral detergent and wipe lightly. 3 Storage • • • • 4 Store the product in a dry place where mold is not likely to form. Store the objectives and eyepieces in a desiccator or similar container with a drying agent. Put a vinyl cover over the product to protect it from dust. Before putting on the vinyl cover, turn off the power switch of the microscope system (press the “ ” side) and wait until the lamp house gets cool sufficiently. Regular Inspections Periodical inspections of the product are recommended in order to maintain peak performance.
